SRX24669104: mRNA sequencing of Variovorax paradoxus or Bacillus mycoides exposed to plant or scrambled miRNA after 20 or 120 minutes

Design: 1 mL of every culture was sampled and centrifuged at 5000 rpm for 5 min. The bacterial pellet was resuspended in 100 L of a lysozyme solution: 1 mg/mL for V. paradoxus (Gram -) and 3 mg/mL for B. mycoides (Gram +). After vortexing the resuspension, the samples were left to incubate for 15 min at room temperature. The following steps used solutions from the RNeasy Plant Mini Kit (QIAGEN). Total RNA was sent for sequencing at the Centre dexpertise et de services Gnome Qubec (Montreal, Canada), where ribosomal RNAs were depleted and messenger RNAs were sequenced.
